The way I would do it, would rbe to put the either oak accorns and or chestnuts in the tub. Prior to placing them in the tub, I would give them a quick wash either in alcohol, bleach or peroxide. Would wait for plants to germinate and put the roots in the substrate. That would be good enough to confirm that the roots are mycorized. Next step would be moving outside and transplanting the young seedlings to the tubs with soil.

Good afternoon . What do you think about replacing petri dishes and test tubes with glass flat-walled bottles? It is essentially a hybrid of a test tube and a petri dish. The only thing that worries me is the convenience of isolating the mycelium strain in a bottle. I would like to hear your opinion.

After isolating a haploid, and growing it out on agar, and combining it with another haploid, will the entire monokaryon become a dikaryon? Or only where they fuse together? Can you make a transfer from the farthest side and it still be monokariotic or does information travel through and know it has mated?

I would only suggest buying blocks to try out the growing process to experiment with. It’s not good to do if making money is your aim.

great video. I have a few questiosn though. Is the purpose of puling ot a syringe full of LC then squirting it back in too mix up the culture? AT time 7:44 when you tip the jar over to fill the syringe alot of liquid drips out onto the table; why, what, and from where?

What would the expected genetic outcome be from spores harvested from a specific phenotype? For example, would M3 spores produce only M3, or all phenotypes?

I used the same syringe filters for gas exchange, but I found that the inoculation was much slower than micropore tape.. and in the case of larger jars, the mycelium would struggle to get to the bottom of the jar. It could be that I got dud filters.. But I also noticed that you have smaller jars. Have you face this issue (which is maybe why you are using these lids with smaller jars?)

@ 10:23 That obnoxious bend in the tubing ends in the middle of the jar so you'd essentially only be able to draw half of your LC through the tubing. You could get the last half with a syringe but that defies the whole point of using the tubing imo

Love your video. May I ask if i do a indoor shop for msuhroom cultivation, i will have a grwoing room in the shop. Do I have to bring air from outdoor dirctly or I can bring air from yhe shop. Becasue i am in canada, winter here is quite cold. How should i do with air exchange? Thank you!

Thank you, spray bottle idea is a gem with the isopropyl alcohol. Growing these things can be tough simply cause how easy contamination occurs
